Feasibility Check & Project Consultion

The feasibility study involves a close dialogue between the customer and our cell culture experts. The customer's requirements are discussed and finally our cell culture experts will suggest you one or more reasonable medium compositions.

Once the customized quotation has been prepared by our team and accepted by the customer, the project can begin.

QC of Incoming Cell Line

During this step of the project we will confirm the culture conditions of the external cell line including mycoplasma test. This step is not necessary when a trenzyme in-house cell line will be used for the project.

  • Thawing and cultivation of cells
  • Thaw-viability assay, microscopic evaluation and mycoplasma test
  • Verification of culture conditions
  • Cryopreservation of a small cell bank

Cell Line Adaption

Depending on the cell line that should be adapted it can be beneficial to perform a stepwise adaption or to perform a so-called “do-or-die”-approach.

During adaption the growth of your cell line will be monitored and documented. You will receive full documentation and medium composition in a comprehensive data sheet or optionally a final report. Viability and growth rates will be monitored and documented over several passages for both approaches

  • Step-Wise Adaption: The old medium will be replaced slowly in several steps (e.g. 25%, 50%, 75%, 100%)
  • „Do-or-Die“-Approach: The old medium will be replaced in one step at once

Cell Line Expansion & Cryopreservation

The adapted cells will be maintained in culture for a few passages in order to validate successful adaption to the new medium. After validation we will expand the cells and cryopreserve them according to your needs (freezing media, number of cells / vial, number of vials).

Quality Control of the Adapted Cell Line

We thaw your newly adapted cryopreserved cell line to scrutinise viability of the cell line. In order to verify the culture conditions we will passage the cells for another three times.

Timeline

  • The timeline very much depends on the cell line, the doubling rate of the cells and on the type of medium (low or high nutrient content) to which the cells shall be adapted. Therefore timeline ranges from 6 to 12 weeks.
